研究方向:天然药物生物智造

长期致力于天然药物生物智造领域的教学与科研,建立了天然产物“新分子、新反应、新药物”上下贯通的“天然药物生物智造”研究新范式,为天然产物“出新率低、成药性差、合成效率低”研究瓶颈的突破提供解决方案。

(1)新分子:建立基于靶标和表型的人工智能辅助“从头药物设计”方法,通过对已知药物的靶向特异性、安全性、药代动力学等药学性质的深度学习,迭代设计符合临床患者需求的候选药物,提高新药研发的成功率。

(2)新反应:人工智能辅助设计人工产物生物合成途径,探究非天然供体引入及其手性控制的酶学机理,建立目标产物体外、体内人工生物合成途径,实现目标产物的“从无到有”。阐明人工生物合成途径与底盘细胞的适配机制,构建合成效率显著提升的细胞工厂,实现目标产物的“从低到高”。

(3)新药物:以临床价值为导向的新药创制为目标,将细胞毒类药物改造为肿瘤特异性靶向候选药物,突破细胞毒类药物毒性大、靶向性差、药效单一等成药难的瓶颈,开发具有全新抗肿瘤机制的原创新药。
